Armenian midfielder Henrikh Mkhitaryan gave Manchester United the lead on a goal assisted by Zlatan Ibrahimovic in the 35th minute.
Eight minutes into the second half Rostov equalized through Aleksandr Bukharov.
Iranian striker Sardar Azmoun came on for Bukharov in the 74th minute but Rostov were unable to find the go-ahead score.
The return leg will be played on 16 March in Old Trafford.
In Piraeus, it was a battle between Greece and Turkey as Olympiacos hosted Turkish giants Besiktas.
Karim Ansarifard was in the starting lineup for Olympiacos and the striker from Ardabil provided the assist for Argentinian midfielder Esteban Cambiasso in the 36th minute.
Besiktas equalized through Cameroonian striker Vincent Aboubakar eight minutes after the restart.
The return leg will be played in Istanbul on 16 March.
